Investigadores portugueses, em colaboração com a Universidade de Barcelona, desenvolvem novos compostos semissintéticos de origem natural com atividade antitumoral notável

O celastrol é um dos triterpenóides naturais com maior atividade antitumoral. Neste trabalho, um conjunto de novos derivados do celastrol foram sintetizados e a sua atividade foi testada em linhas celulares tumorais humanas. Entre os novos compostos sintetizados, muitos apresentaram um efeito de inibição do crescimento celular mais acentuado do que o próprio celastrol. O composto 24 foi o que mais se destacou, exibindo uma atividade antiproliferative excecional e também uma melhoria importante da seletividade em células tumorais versus células não-tumorais. O mecanismo molecular deste composto foi estudado mais profundamente em linhas celulares tumorais humanas do ovário (SKOV-3). Os resultados demonstraram que o composto 24 induziu a morte celular através da via extrínseca da apoptose. Outros mecanismos moleculares foram explorados, revelando resultados interessantes a nível de proteínas específicas como a p53, Hsp90 entre outras, nomeadamente na via Akt/mTOR. Destaca-se ainda o efeito sinergético deste novo composto com a cisplatina nas células SKOV-3. Estes resultados, em conjunto, sugerem que o composto 24 poderá ser um lead promissor para o desenvolvimento de novas terapias antitumorais.

Abstract:

Celastrol is one of the most active antitumour compounds among the natural triterpenoids. A series of new celastrol derivatives have been synthesised and analysed for their biological activity against human cancer cell lines. Several compounds presented a stronger growth inhibition effect than celastrol on the cell lines studied. Among them, compound 24 was the most promising derivative, as it exhibited both a remarkable antiproliferative activity and an improved selectivity in tumour versus non-tumour cells. The anticancer molecular mechanism of compound 24 in the human ovary cancer cell line SKOV-3 was further studied and the results showed that compound 24 induced apoptosis through the activation of the extrinsic death receptor pathway. Interestingly, the results revealed that compound 24 might be able to decrease the levels of dysfunctional p53. The assays also suggested that compound 24 is an Hsp90 inhibitor, and that the Akt/mTOR pathway might be involved in the downstream regulation that leads to its antiproliferative activity. Moreover, a synergistic anticancer effect was evidenced when SKOV-3 cells were simultaneously treated with compound 24 and cisplatin. Taken together, these results suggest that compound 24 may be a promising lead for the development of new cancer therapies.
