Disruption of breast cancer cell pH dynamics decreases aggressiveness and potentiates response to chemotherapy

The reverse pH gradient is a common feature of cancer cells, in which the intracellular pH is more alkaline and the extracellular more acidic. This phenotype is supported by increased activity of pH regulators like ATPases, carbonic anhydrases (CAs), monocarboxylate transporters (MCTs) and sodium–proton exchangers (NHEs). In this work, we analyzed the expression of these pH regulators and explored their inhibition in breast cancer cells as a therapeutic strategy.

O gradiente reverso de pH é uma característica comum das células malignas, em que o pH intracelular é mais alcalino e o extracelular mais acídico, fator que está associado a uma maior agressividade. As células malignas conseguem manter este gradiente de pH à custa da atividade aumentada de várias proteínas reguladoras de pH (proteínas membranares), como sejam as ATPases, anidrases carbónicas (CAs), transportadores de monocarboxilatos (MCTs) e os trocadores sódio–protão (NHEs).

The Adaptive Immune Landscape of the Colorectal Adenoma–Carcinoma Sequence

The tumor immune microenvironment exerts a pivotal influence in tumor initiation and progression. The aim of this study was to analyze the immune context of sporadic and familial adenomatous polyposis (FAP) lesions along the colorectal adenoma–carcinoma sequence (ACS). We analyzed immune cell counts tumor mutation burden, MHC-I expression and PD-L1 expression.  The colorectal ACS is characterized by a progressive loss of adaptive immune infiltrate and by the establishment of a progressively immune cold microenvironment.

Modulação do sistema imunitário adaptativo na sequência adenoma-carcinoma num contexto de tumores colo-retais

O sistema imunitário tem um papel fundamental na iniciação e progressão do tumor. A interação entre tumores, quer hereditários quer esporádicos, e o sistema imunitário pode variar ao longo da progressão tumoral. Neste artigo fez-se uma análise das células imunitárias no microambiente tumoral, tal como de marcadores importantes para o sistema imunológico em displasias de baixo grau, alto grau e adenocarcinomas colo-retais.
